There are two border-crossings between Macau and Mainland China: the Barrier Gate and the COTAI Frontier Post. The Barrier Gate operates from 7:00 AM to midnight, while the COTAI Frontier Post, at the Taipa-Coloane Reclaimed Area, operates from 9:00 AM to 8:00 PM.
Kee Kwan Motor Road Co. offers an air-conditioned bus service between Macau and Guangzhou, as well as to other major cities in Guangdong Province. Buses to Guangzhou depart once every hour from the bus stop near the Master Hotel (on Rua das Lorchas, near the end of Avenida de Almeida Ribeiro). Buses to Guangzhou also depart from Macau airport.

China Travel Services operates a large number of daily cross-border buses, from Hong Kong. Using them is a good option, both because the fare is significantly lower than the train and also because they pick their passengers from different points in Hong Kong and drop them off in various points in Shenzhen and Guangzhou (saving you the hassle of getting to and from the railway station).
One way fare from Hong Kong to Guangzhou is around HK$ 100
One way fare from Hong Kong to Shenzhen is around HK$ 60
The company also operates direct bus line from Hong Kong International Airport to the large cities on the Chinese side of the border.
